We uninstalled Nospam and now are getting no response from the SMTP server. Within Exchange SMTP Virtual server, we reset the port to the default. Any suggestions of where else to make changes?

have you verified that the exchange server is indeed listen on 25 now? get the freeware tool called aports to view the active ports on your server to verify open ports
